The β-galactosidase gene (8 amino acids deleted from N-terminus) was cloned into one of the Avidity N-terminus vectors (PAN5) creating a fusion protein of AviTag-β-gal. The plasmid can be transformed into bacterial strain background of interest by conventional means. The strain can then be induced in parallel with AviTag-protein of interest according to the following
induction procedure
. The following procedure is for AviTagged-protein in the Avidity strain AVB101. AVB101 contains the chloramphenicol resistant plasmid pACYC-184 that overexpresses the Biotin Ligase, BirA. If your strain is different, just disregard the use of chloramphenicol.
